Traditional Polymerase Chain Reactions (PCR) detect the amplified DNA product, or amplicon, in an end-point analysis. This is done by visualizing the DNA product on an agarose Gel following completion of the reaction. The number of amplification cycles required for the reaction to be successful. However, real-time PCR allows for simultaneous evaluation of the progress of the reaction and the accumulation of amplification products. After each cycle, product quantification can be done in real-time. Digital PCR (dPCR), an improvement on traditional PCR methods, is used to directly quantify and clonally amplify nucleic acid such as DNA, DNA, RNA or methylated DNA. Digital PCR is still in its early stages. The method used to measure the nucleic acid amounts is what makes dPCR different from conventional PCR. The sensitivity of dPCR is higher than qPCR and is therefore more suitable for applications with low target sequences. The technology can detect mutation genes on a low scale in rare diseases and HIV DNA in cells of patients.
